I have a WD external hard disk and has 146GB of free space. I wanted to copy a 4GB iso file into it and an error popped out saying "The file '~file name~.iso' is too large for the destination file system".
Can anyone tell me why is this happening??? Many thanks.

This is mostly likely because your external drive is formatted to FAT32 system, which has a file size limit of 4GB.
You could convert your drive or partition an NTFS system.
But the easiest thing if you don't need to run this ISO directly from the drive is to compress it and or break it up into smaller files. You can use any zip or other archiving tool, but it might not compress the file size any smaller, but most of these tools should also be able to split an archived file into smaller files. When you need it again, it should be an easy command to merged them back together again off the external drive when you need to use the file.

It might be due to the HDD format.. Look at the Properties tab of the external hard drive, then look under the General tab at the file system. If it says FAT or FAT32, this might be the reason. As a FAT or a FAT32 files system have a max file size of 4gig, where as a NTSF file system can have any file size...
